چکیده
Inflammation of the adipose tissue contributes to onset and progression several chronic obesity-related diseases. The two most important lipophilic diterpenoid compounds found in root Salvia milthorrhiza Bunge (also called Danshen), tanshinone IIA (TIIA) cryptotanshinone (CRY), have many favorable pharmacological effects. However, their roles obesity-associated adipocyte inflammation related sub-networks not been fully elucidated. In present study, we investigated gene, miRNAs protein expression profile prototypical dysfunction markers inflamed human adipocytes treated with TIIA CRY. results showed that CRY prevented tumor necrosis factor (TNF)-α induced inflammatory response adipocytes, by counter-regulating pattern secreted cytokines/chemokines associated (CCL2/MCP-1, CXCL10/IP-10, CCL5/RANTES, CXCL1/GRO-α, IL-6, IL-8, MIF PAI-1/Serpin E1) via modulation gene (as demonstrated for CCL2/MCP-1, IL-8), as well miRNA (miR-126-3p, miR-223-3p, miR-124-3p, miR-155-5p, miR-132-3p), attenuating monocyte recruitment. This is first demonstration a beneficial effect on obesity development complications, offering new outlook prevention and/or treatment metabolic

Metabolism of tanshinone IIA, cryptotanshinone and tanshinone I from Radix Salvia miltiorrhiza in zebrafish.
The study aimed to investigate the potential of zebrafish in imitating mammal phase I metabolism of natural compounds. Three diterpenoid quinones from Radix Salvia miltiorrhiza, namely tanshinone IIA (TIIA), cryptotanshinone (Cry) and tanshinone I (TI) were selected as model compounds, and their metabolites mediated by zebrafish were characterized using a high-performance liquid chromatography ...
متن کاملInflammatory Gene Expression Patterns Revealed by DNA Microarray Analysis in TNF-α-treated SGBS Human Adipocytes
We report here the use of human inflammation arrays to study the inflammatory gene expression profile of TNF-alpha- treated human SGBS adipocytes. Human preadipocytes (SGBS) were induced to differentiate in primary culture, and adipocyte differentiation was confirmed, using Oil Red O staining. We treated the differentiated adipocytes with TNF-alpha, and RNA from differentiated adipocytes with o...
متن کاملTanshinone IIA attenuates ovalbumin-induced airway inflammation and hyperresponsiveness in a murine model of asthma
Objective(s): Tanshinone IIA (T. IIA), one of the most pharmacologically active components extracted from Salviae miltiorrhiza, has anti-inflammatory and antioxidant features. The aim of the present study is to investigate the benefit of T. IIA on asthma using a murine model of asthma induced by ovalbumin (OVA). Materials and Methods: Male BALB/c mice were used in the present study. The mice we...
متن کاملIsolation of Tanshinone Iia and Cryptotanshinone in Salvia Miltiorrhiza Using Two Conventional Extraction Techniques and Quantification by Validated Hplc Method
This study describes a simple, easy and inexpensive method for extracting tanshinone IIA and cryptotanshinone from Salvia miltiorrhiza by sonication or decoction. A comparison of sonication and decoction showed a similar yield for cryptotanshinone, while the tanshinone IIA yield was slightly higher using sonication. An analytical method for detecting tanshinone IIA and cryptotanshinone in extra...
متن کاملMetformin and Pioglitazone Reduce Gene Expression of Inflammatory Factors in Insulin Resistant and Hypertrophied Adipocytes
Objective: In obesity, chronic low grade inflammation, created by induction of pro-inflammatory markers, causes adipocyte dysfunction in adipose tissue. Adipocytes dysfunction is associated with various diseases including insulin resistance and obesity. In obesity, inflammatory factors such as osteopontin (OPN), angiopoietin-like protein 2 (Angptl2) and transforming growth factor-β (TGF-β) are...
